    Genomic analysis of early murine mammary gland development using novel probe-level algorithms

    We describe a novel algorithm (ChipStat) for detecting gene-expression changes utilizing probe-level comparisons of replicate Affymetrix oligonucleotide microarray data. A combined detection approach is shown to yield greater sensitivity than a number of widely used methodologies including SAM, dChip and logit-T. Using this approach, we identify alterations in functional pathways during murine neonatal-pubertal mammary development that include the coordinate upregulation of major urinary proteins and the downregulation of loci exhibiting reciprocal imprinting

    An Outbreak of Severe Infections with Community-Acquired MRSA Carrying the Panton-Valentine Leukocidin Following Vaccination

    Background: Infections with community-acquired methicillin-resistant Staphylococcus aureus (CA-MRSA) are emerging worldwide. We investigated an outbreak of severe CA-MRSA infections in children following out-patient vaccination. Methods and Findings: We carried out a field investigation after adverse events following immunization (AEFI) were reported. We reviewed the clinical data from all cases. S. aureus recovered from skin infections and from nasal and throat swabs were analyzed by pulse-field gel electrophoresis, multi locus sequence typing, PCR and microarray. In May 2006, nine children presented with AEFI, ranging from fatal toxic shock syndrome, necrotizing soft tissue infection, purulent abscesses, to fever with rash. All had received a vaccination injection in different health centres in one District of Ho Chi Minh City. Eight children had been vaccinated by the same health care worker (HCW). Deficiencies in vaccine quality, storage practices, or preparation and delivery were not found. Infection control practices were insufficient. CA-MRSA was cultured in four children and from nasal and throat swabs from the HCW. Strains from children and HCW were indistinguishable. All carried the Panton-Valentine leukocidine (PVL), the staphylococcal enterotoxin B gene, the gene complex for staphylococcal-cassette-chromosome mec type V, and were sequence type 59. Strain HCM3A is epidemiologically unrelated to a strain of ST59 prevalent in the USA, althoughthey belong to the same lineage. Conclusions. We describe an outbreak of infections with CA-MRSA in children, transmitted by an asymptomatic colonized HCW during immunization injection. Consistent adherence to injection practice guidelines is needed to prevent CA-MRSA transmission in both in- and outpatient settings

    OBJECTIVES:Articular cartilage damage related to irreversible physical disability affects most patients with chronic rheumatoid arthritis (RA). Strategies targeting the preservation of cartilage function are needed. Laser acupuncture (LA) can be an emerging alternative therapy for RA; however, its molecular mechanism underlying the beneficial effect on cartilage has not been elucidated. This study aimed to examine the potential chondroprotective effects of LA on extracellular matrix (ECM) macromolecules and proinflammatory cytokines in the articular cartilage of adjuvant-induced arthritis (AIA) rats and explore its related mechanisms. DESIGN:Monoarthritis was induced in adult male Sprague-Dawley rats (250-300 g) via intraarticular injection of complete Freund's adjuvant (CFA) into the tibiotarsal joint. Animals were treated with LA at BL60 and KI3 acupoints three days after CFA administration with a 780 nm GaAlAs laser at 15 J/cm2 daily for ten days. The main outcome measures including paw circumference, paw withdrawal threshold, histopathology and immunoassays of tumor necrosis factor-α (TNF-α), collagen type II (CoII), cartilage oligomeric matrix protein (COMP) were analyzed. RESULTS:LA significantly reduced ankle edema and inflammation-induced hyperalgesia in AIA rats (P < 0.05). Moreover, the TNF-α levels were significantly decreased while CoII, COMP and proteoglycans proteins were significantly enhanced following LA stimulation of the AIA cartilage compared to those treated with sham-LA (P < 0.05). CONCLUSIONS:LA attenuates cartilage degradation in AIA rat by suppressing TNF-α activation and up-regulating ECM macromolecules, suggesting LA might be of potential clinical interest in RA treatment

    Impact of Phase II cardiac rehabilitation on abnormal heart rate recovery

    Background: A delayed decrease in heart rate recovery (HRR) following a maximal exercise test is a predictor of mortality in healthy adults and in those referred for diagnostic testing evaluation. Cardiac rehabilitation (CR) has been proven to be beneficial in the treatment of numerous diseases. The purpose of this retrospective study was to explore the relationship between CR and improvement of HRR and to determine which factors can be predictive for the improvement of abnormal HRR after Phase II CR. Methods: We reviewed the medical histories of patients referred for symptom-limited cardiopulmonary treadmill testing with abnormal HRR. Each patient underwent exercise testing before and after Phase II CR, and parameters including functional aerobic impairment, metabolic equivalent, anaerobic threshold (AT), and %AT were recorded and compared. Results: Of the 25 patients recruited, 13 patients showed no abnormal HRR after Phase II CR. The functional capacity of these patients was improved after Phase II CR, to a statistically significant extent; the aerobic capacity (as expressed with AT and %AT) showed improvement, but without statistical significance. Conclusion: There are multiple factors of cardiopulmonary exercise tests that cannot be used to predict the effect of Phase II CR on the improvement of abnormal HRR. Forty-one percent of patients with abnormal HRR could improve after Phase II CR, but all of the patients could have improved exercise capacity regardless of whether or not HRR improved. We can conclude that HRR and exercise capacity change independently. However, it is important to closely follow-up during Phase III CR for patients with persistently abnormal HRR after Phase II CR has been completed

    Semantic-Event Based Analysis and Segmentation of Wedding Ceremony Videos

    Wedding is one of the most important ceremonies in our lives. It symbolizes the birth and creation of a new family. In this paper, we present a system for automatically segmenting a wedding ceremony video into a sequence of recognized wedding events, e.g., the couple’s wedding kiss. Our goal is to develop an automatic tool for users to efficiently organize, search, and retrieve his/her treasured wedding memories. Furthermore, the event descriptions could benefit and complement the current research in semantic video understanding. Technically, three kinds of event features, i.e., the speech/music discriminator, flashlight detector, and bride indicator, are exploited to build statistical models for each wedding event. Events are then recognized by a hidden Markov model, which takes into account both the fitness of observed features and the temporal rationality of event ordering to improve the segmentation accuracy. We conducted experiments on a rich set of wedding videos, and the results demonstrate the effectiveness of our approach
